照例去了海边，照例吃了月饼，不过超级想念当年在清华时老妈给带的那箱月饼，真是活生生吃了好几个月，这一晃都过去7年了，岁月真TM催人老。什么时候能在家过次中秋节啊！~~~~

An earthquake measuring 7.8 on the Richter scale struck the Sichuan Province in China on May 12 at 2:28 pm local time. So far the death toll stands at 12,000 and the numbers are expected to rise sharply as rescuers approach the epicenter. 60,000 people are still unaccounted for, either missing or trapped under the debris. Tens of thousands of homes, factories, and offices have collapsed which killed many instantly, while burying others. The earthquake was so powerful that it sent people rushing out of buildings a thousand miles away in Beijing and Shanghai. We will set up a booth on UCI campus to receive your donation:

A key role for diacylglycerol lipase-alpha in metabotropic glutamate receptor-dependent endocannabinoid mobilizationKwang-Mook Jung 1, Giuseppe Astarita 1, Chenggang Zhu 1, Matthew Wallace 2, Ken Mackie 2, Daniele Piomelli 3*1 University of California, Irvine 2 University of Washington 3 Univ. of California - Irvine * Address correspondence to: E-mail: piomelli@uci.edu Activation of group-I metabotropic glutamate (mGlu) receptors recruits the endocannabinoid system to produce both short- and long-term changes in synaptic strength in many regions of the brain. Although there is evidence that the endocannabinoid 2-arachidonoylglycerol (2-AG) mediates this process, the molecular mechanism underlying 2-AG mobilization remains unclear. In the present study, we used a combination of genetic and targeted lipidomic approaches to investigate the role of the postsynaptic membrane-associated lipase, diacylglycerol lipase type-alpha (DGL-alpha), in mGlu receptor-dependent 2-AG mobilization. DGL-alpha overexpression in mouse neuroblastoma Neuro-2a cells increased baseline 2-AG levels. This effect was accompanied by enhanced utilization of the 2-AG precursor 1-stearoyl,2-arachidonoyl-sn-glycerol and increased accumulation of the 2-AG breakdown product arachidonic acid. A similar, albeit less marked response was observed with other unsaturated and polyunsaturated monoacylglycerols, 1,2-diacylglycerols and fatty acids. Silencing of DGL-alpha by RNA interference elicited opposite lipidomic changes to those of DGL-alpha overexpression, and abolished group-I mGlu receptor-dependent 2-AG mobilization. Coimmunoprecipitation and site-directed mutagenesis experiments revealed that DGL-alpha interacts, via a PPxxF domain, with the coiled-coil (CC)-Homer proteins Homer-1b and Homer-2, two components of the molecular scaffold that enables group-I mGlu signaling. DGL-alpha mutants that do not bind Homer maintained their ability to generate 2-AG in intact cells, but failed to associate with the plasma membrane. The findings indicate that DGL-alpha mediates group-I mGlu receptor-induced 2-AG mobilization. They further suggest that the interaction of CC-Homer with DGL-alpha is necessary for appropriate function of this lipase.
October 16 Brake ChangeDashboard上的小黄灯实在看不下去了，特别是每次踩刹车时听到撕心裂肺的磨擦声之后，终在网上订了Part：brake pad set, pad sensor和brake disc.并且在达人的帮助下，于周日在一片空旷的parking lot中大干一场，终于把PAD换掉了。其实步骤不难：撑车-卸轮子-卸刹车-装刹车-装轮子，换边，再搞一动。详细说来，撑车不难，是人都会，可还是把手给擦了；卸轮子也容易，没有电动的就裸搞。可是到刹车就难了，按理说有个CLIP，打开就能把旧的PAD卸下来，把新的装上，就OK了，可是BIMMER要把整个刹车给下下来，才能把旧的PAD卸下来。可是卸的时候千万不能碰到Brake Fluid的管子，很容易搞废的，费了九牛二虎之力，终在工具不足的情况下，成功地拆下两个固定螺丝，装新的刹车片装好，再原样组装起来。而且把pad sensor也换了新的，小灯终于不亮了。本来也要换PAD的，但达人说没必要，还挺平的，就这么搞定收工了。
PS，装轮胎的时候五个螺钉要走五角星的线路，不然开起来会晃。 |
